ABSTRACT

Background@#Fatty acid-binding protein 4 (FABP4) has been demonstrated to be a predictor of early diabetic nephropathy. However, little is known about the relationship between FABP4 and diabetic retinopathy (DR). This study explored the value of FABP4 as a biomarker of DR in patients with type 2 diabetes mellitus (T2DM). @*Methods@#A total of 238 subjects were enrolled, including 20 healthy controls and 218 T2DM patients. Serum FABP4 levels were measured using a sandwich enzyme-linked immunosorbent assay. The grade of DR was determined using fundus fluorescence angiography. Based on the international classification of DR, all T2DM patients were classified into the following three subgroups: non-DR group, non-proliferative diabetic retinopathy (NPDR) group, and proliferative diabetic retinopathy (PDR) group. Multivariate logistic regression analyses were employed to assess the correlation between FABP4 levels and DR severity. @*Results@#FABP4 correlated positively with DR severity (r=0.225, P=0.001). Receiver operating characteristic curve analysis was used to assess the diagnostic potential of FABP4 in identifying DR, with an area under the curve of 0.624 (37% sensitivity, 83.6% specificity) and an optimum cut-off value of 76.4 μg/L. Multivariate logistic regression model including FABP4 as a categorized binary variable using the cut-off value of 76.4 μg/L showed that the concentration of FABP4 above the cut-off value increased the risk of NPDR (odds ratio [OR], 3.231; 95% confidence interval [CI], 1.574 to 6.632; P=0.001) and PDR (OR, 3.689; 95% CI, 1.306 to 10.424; P=0.014). @*Conclusion@#FABP4 may be used as a serum biomarker for the diagnosis of DR.

ABSTRACT

Objective To study the predictive value of platelet index on hemodynamically significant patent ductus arteriosus (hsPDA) in preterm infants.Method The data of 120 preterm infants born between January 2015 and December 2016 were retrospective analyzed,including 60 cases of non-hsPDA (nhsPDA) group and 60 cases of hsPDA group.The clinical features and platelet associated indicators between nhsPDA group and nhsPDA group were compared.Multivariate Logistic regression was used to analyze the effects of various factors on the occurrence of hsPDA.Receiver operating characteristic (ROC) curve was used to evaluate the early predictive value of platelet associated indicators for hsPDA.Result The hsPDA group had statistically significant differences in many parameters (P < 0.05) comparing with the nhsPDA group,including smaller gestational age,lower birth weight and Apgar's score (1 min and 5min),lower proportion of cesarean section and preeclampsia,lower mean platelet volume and platelet distribution width (PDW),bigger PDA diameter and diameter2/birth weight,more serious respiratory distress syndrome,and a higher rate of pulmonary surfactant use.Multivariate Logistic regression analysis indicated that the risk of hsPDA was increased as the PDW decreased (OR =1.240,95% CI 1.011 ~ 1.521).The ROC curve analysis showed that the best diagnostic value of PDW was 12.9%,and the sensitivity of early prediction of hsPDA was about 53.3%,and the specificity was 78.3%.Conclusion PDW has certain early predictive value for hsPDA in preterm infants.Preterm infants with PDW < 12.9% need to be monitored closely for the occurrence of hsPDA.

ABSTRACT

Objective To compare the clinical effects of 577 nm and 532 nm laser panretinal photocoagulation in the treatment of severe non-proliferative diabetic retinopathy (NPDR).Methods A prospective,controlled trial was conducted in 42 patients(64 eyes)with severe NPDR,who were randomly divided into 577 nm group and 532 nm group.All of patients received PRP with the single-point model.Preoperative and postoperative 1 day,1 month,3 and 6 months,the best corrected visual acuity (BCVA),fundus,optical coherence tomography (OCT) and full field flash electroretinogram (F-ERG) were examined.After treatment 3 and 6 months,fundus fluorescein angiography (FFA) examination was performed between two groups.Results In 577 nm group and 532 nm group,the average number of laser spot was (1969.25 ± 278.19) and (2098.16 ± 289.27) respectively;average laser power was (425.23 ± 50.15)mW and (438.15 ± 38.48)mW respectively;and average energy density was (7.54 ± 1.54) mW · ms · μm-2 and (7.68 ± 3.01)mW · ms · μm-2.There was no difference in number of laser spot(t =2.68),laser power (t =1.46) and energy density (t =2.15) between the two groups (all P > 0.05),and the differences of macular central thickness after treatment 1 month,3 and 6 months (t =1.98,1.88,1.81 respectively) approached no statistical significance between the two groups (all P > 0.05),while F-ERG a,b wave amplitudes after treatment 1 month,3 and 6 months (a wave:t =5.94,5.19,6.97;b wave:t =5.67,4.56,5.12) had significant differences between groups (all P < 0.05).The effective rate of treating 6 months after operation in the two groups were 87.5% and 46.9% respectively,with significant difference (x2 =7.56,P < 0.05).Conclusion 577 nm laser is more effective and has less damage to visual function than 532 nm laser in the treatment of NPDR.

ABSTRACT

Objective To optimize the processing condition of the extraction of Baphicacanthis cusiae ( Nees) Bremek. roots polysaccharides ( BCP) by ultrasound technology, and to provide the basis for the further development and study of BCP. Methods The extraction temperature, extraction time, liquid-solid ratio were used as the influence factors, and the yield of polysaccharides from Baphicacanthis cusiae roots was used as the evaluation index. On the basis of single-factor test, 3-factor and 3-level Box-Behnken test was designed for quadratic polynomial regression equation of the yield of polysaccharides. And then the response surface methodology was used for the optimization of process condition. Results The optimum extraction condition was as follows: extraction temperature was 60℃, extraction time was 35 min, and liquid-solid ratio was 24.5∶1 (V∶m, mL·g-1). Under the optimal condition, the yield of polysaccharides was 83.7 mg·g-1. Conclusion The method is simple, rapid and highly efficient, which will provide experimental basis for the development and application of Baphicacanthis cusiae roots polysaccharides.

ABSTRACT

This retrospective analysis showed that the most frequent pathogen causing bacterial liver abscess was Klebsiella pneumoniae in 138 patients.Compared with the patients without diabetes mellitus,it was found that:( 1 ) the percentage of diabetic patients having typical abdominal pain was lower ( P < 0.05 ) ; ( 2 ) neutrophilic granulocytosis was more marked,but albumin and hemoglobin levels were lower in diabetic patients( P<0.05 ) ; ( 3 )more diabetic patients were complicated with urinary tract infection and suffered from septicemia( P<0.05 ) ; (4) the clinical course of treatment in diabetic patients was much more prolonged( P<0.05 ).

ABSTRACT

Objective To identify synovium-associated proteins of by analyzing proteome of FLS between RA patients and traumatic arthritis. Methods The total protein of FLS from 3 patients of RA and 6 normal controls were separated by 2-DE. The differentially expressed proteins were identified by MALDI-TOF-MS. Results The 2-DE image results showed that there were many proteins differentially expressed in FLS from RA and control. A total of 33 differential proteins that were up-regulated in RA as compared with controls were selected and 30 proteins including pyruvate kinase isozymes M1/M2, α-enolase, protein disulfide-isomerase A3 precursor, glutathione transferase, peroxiredoxin 1, ubiquitin-conjugating enzyme E2K, platelet-activating factor acetylhydrolase IB, dihydropyrimidinase-related protein 2, adenosine kinase, transaldolase, δ-δ-dienoyl-CoAisomerase, 26S protease regulatory subunit 6A, annexin All, tryptophanyl-tRNA synthetase isoform b, lamin-A/C, myomesin-1 isoform b, ruvB-like 1, actin-cytoplasmic 1, T-complex protein 1, stress-70 protein, mitochondrial precursor, α-crystallin B chain, 78 000 glucose-regulated protein, PSME4, SELENBP1 and so on. Conclusion The differentially expressed proteins are present in FLS from RA and control, which may be the synovium biomakers and involved in pathogenesis of RA.

ABSTRACT

Objective To investigate the relationship between nutcracker syndrome (NCS) and glomerular disease of patients with hematuria and/or proteinuria. Methods To analysis clinical and pathological data of 39 patients with NCS complicated glomerular nephritis. Ultrasonic examination, 24h urinary protein quantization, urine routine, urinary sediment examination and urinary protein electrophoresis were used in all patients .Among them,30 patients accepted renal biopsy. Results Among the 39 patients, hematuria and/or proteinuria were aggravated in most of them after exercise. Multiformity was manifested by renal biopsy. Definite diagnosis can be made through clinical, laboratory examination and renal biopsy. Conclusions NCS could coexist with glomerular disease. NCS patients with hematuria and/or proteinuria should be carried out renal biopsy when needed whether coexisted with glomerular nephritis or not.

ABSTRACT

Objective To explore the mechanism of protection role of ambroxol against acute lung injury (ALI)by studying the change of cytokine mRNA expression during the course of ALI.Method The study was composed of experiments both in vivo and in vitro. (1)Experiments in vivo were as follows.Twenty-four sprague-Dawley rats were randomly divide into 3 groups,namely,control group(n=8),acute lung injury group(LPS group,n=8)and ambroxol group(LPS+A group,n=8).The rat model of acute lung injury was induced by intraperitoneal injection of 10 mmol/L lipoopolysaccharide(LPS).The pathological alteration of lung tissue and arterial partial pressure of oxygen(PaO2)were observed.Expressions of TNF-α,IL-10 and IL-24 mRNA were determined by using RT-PCR assay. (2)Experiments in vitro were the followings.Alveolar macrophage cells were collected and divided into 3 groups as above mentioned.Cells were treated with normal saline,with LPS,and with LPS plus ambroxolin dose of 180 μmol/L at 0,6,12 and 24 hours after exposure of LPS,respectively,in 3 groups as above stated.The expressions of TNF-α,IL-10 and IL-24 mRNA were also determine by using RT-PCR.Results The pathological changes of could be parially ameliorated by giving ambroxol.Massive hemorrhage along with vascular edema and infiltration occurred in the lungs of ALT rats.The pathological alterations in ALI rats treated with ambroxol were less severe.The expressions of TNF-α,IL-10 and IL-24 mRNA were dramatically increased in ALI rats,and were partially attenuated after treatment of ambroxol.Macrophages oxposed to LPS for 6 hours showed dramatical increase in expressions of TNF-α,IL-10 and IL-24mRNA those remained at high levels afterwards.The expressions of TNF-α,IL-10 and IL-24 mRNA in macrophages after exposure to both LPS and ambroxol were increased less than those exposed to LPS alone.Conclusions Ambroxol can partially ameliorate the expressions of TNF-α,IL-10 and IL-24 mRNA in alveolar macrophage induced by LPS.

ABSTRACT

Objective:To investigate the expression of MMP-1 and MMP-8 in periodontal tissues in rats with periodontitis at different stages of inflammation with varied severity. Methods:Periodontitis was induced by silk ligature. P.gingivalis(Pg) or Pg with F.nu were used to induce the varied severity of periodontitis in 40 rats. 14 and 28 days after periodontitis induction the animals were sacrificed, periodontal tissues were immunohistochemically stained by antibody of MMP-1 and MMP-8 respectively. Results:MMP-1 and MMP-8 were both strongly positive in gingival epithelia cells and fibroblasts in periodontal ligament in rats with periodontitis.Higher expression of MMP-1 and MMP-8 was observed at 14 d than at 28 d(P0.05). Conclusion:The expression of MMP-1 and MMP-8 varies in different stage of periodontitis. MMP-1 and MMP-8 may play an important role in development of periodontitis.

ABSTRACT

Objective:To assess the clinical and microbiologic effects of ultrasonic scaling with simultanous irrigation with chlorhexidine(CHX), conventional ultrasonic scaling or subgingival irrigation. Methods: 20 teeth in 7 patients with periodontitis were equally divided into test and control group. Patients in the test group were treated by irrigation with CHX during ultrasonic scaling(Master400),those in the control with irrigating after scaling(MiNi ). Clinical assessments of bleeding on probing (BOP),probing depth (PD) were made just before and 7 and 14 days after treatment. The subgingival microbiologic examination was performed before and 0,7 and 14 days after treatment.
